DA-depleted mice with unilateral lesions of the SNc show relatively normal rates of locomotor activity on an open field, as do WT mice, but show a significant decrease in the rate of spontaneous locomotor activity. Furthermore, when MPTP-treated mice were tested on an accelerating rotarod, those which showed a marked loss of TH-immunoreactive (ir) cells in the SNc showed marked impairment in locomotor activity. The deficit in the ability to produce PPI and the pronounced loss of locomotor activity in MPTP-treated mice were positively correlated.
